Applying proteomics to thyroid tumour diagnosis

The application of proteomics in clinical thyroid logy is innovative and is a novel tool to decipher Pathophysiologie-inherent changes in protein function, which cannot be comprehensively studied by the more widely applied transcriptome methods.

In this talk we will provide an overview on methods of thyroid proteomics and their possible pitfalls, present current studies on this topic and discuss our own experience with the application of proteomics technology to study differential protein expression in benign cold thyroid nodules (CTN).

Thus, using a combined approach of 2D gel-electrophoresis (2D-GE) and mass spectrometry, followed by western blot analysis and immunohistochemical studies, we demonstrate for the first time, that benign CTNs are characterised by the up-regulation of several components of the thyroid hormone synthesis machinery, with a strong induction of the H2O2 generating and detoxifying systems. Moreover, increased formation of 8-oxo-guanidine DNA-adducts was detected in CTNs and could besides the increased cell proliferation in these nodules confer an increased potential for genotoxicity and mutagenesis.
