Influence of the Y chromosome on inter-individual variation in cholesterol serum levels of young men

Males display an increased level of cardiovascular morbidity and mortality than age matched females. Among the genetic factors responsible for the relationship between higher cardiovascular risk and male gender are the sex chromosomes. In this study we investigated the association between serum lipid profile and a polymorphism on the non-recombining region of the human Y chromosome in a population of young Caucasian individuals. 1157 young males were recruited form the general population in the south of Poland. They were phenotyped for blood pressure, weight, height, serum lipid profile, glucose and creatinine levels. DNA from each individual was amplified and genotyped for the HindIII (-/+) marker within the non-recombining region of the Y chromosome by RFLP. There was no statistical difference in age, BMI and blood pressure between both genotypes. The HindIII(-) polymorphism compared with the HindIII(+) variant was associated with increased total cholesterol (4.3+0.9 mmol/L vs 4.1+0.8 mmol/L, P=0.01, CI=4.2-4.4), and low-density-lipoprotein cholesterol (LDL-C) (2.6+0.8 mmol/L vs 2.4+0.8 mmol/L, P=0.005, CI=2.53-2.65). The number of individuals with the HindIII(-) genotype increased [from 23% to 27%] across the percentile distribution of serum cholesterol levels, and was accompanied by decreasing number of individuals [from 28% to 20%] with the HindIII(+) genotype (P=0.01). Genetic variation on the Y chromosome may therefore contribute to inter-individual variation in lipid levels.
