Medical treatment in adrenal insufficiency: Does the patient’s quality of life differ depending on which glucocorticoid is given?

Bleicken Benjamin , Ventz Manfred , Strasburger Christian J , Quinkler Marcus

Objective: Patients from FRG and West-Berlin with primary (1AI) or secondary (2AI) adrenal insufficiency were treated with hydrocortisone (HC) while in contrast patients from the former GDR received prednisolone (PR). Investigation of our study is to detect any differences in quality of life (QoL) in patients on HC or PR medication. Methods: 350 patients of our outpatient clinic were contacted by mail. Up to now 131 patients took part in the survey. 2 sta...

17β-estradiol regulates pituitary AnxA1 expression and externalization in vivo and in vitro

Davies EL , Christian HC , Omer S , Buckingham JC , Morris JF

Annexin A1 (AnxA1) acts as a mediator of glucocorticoid (GC) actions in neuroendocrine systems. In the anterior pituitary Anx-1 is expressed mainly by folliculostellate (FS) cells and mediates the early-delayed feedback inhibition exerted by GCs on release of ACTH and other pituitary hormones. GC cause the externalization of AnxA1. The stress responsiveness of the female rat hypothalamo-pituitary-adrenal system varies with the estrous cycle, with increased CORT release in resp...

Structural analysis of somatotrophs and lactotrophs in annexin 1 null mice

Afzal T , Morris JF , Buckingham JC , Christian HC

Annexin 1 (ANXA1) is a member of the annexin family of phospholipid- and calcium- binding proteins with a well-demonstrated role in the inhibitory actions of glucocorticoids on the release of ACTH, TSH, GH and PRL from the anterior pituitary. Consistent with these findings plasma PRL content is raised in male ANXA1 null mice vs wild-type controls (Cover et al Endocrine Abstracts 8, P69). Here we test the hypothesis that because ANXA1 mediates some of the inhibitory actions of ...

Effects of targeted ablation of GHRH neurons in mice on anterior pituitary somatotrophs and lactotrophs

Miller A , Le Tissier P , Robinson I , Christian HC

Animal and clinical models of GHRH excess suggest that GHRH provides an important trophic drive to pituitary somatotrophs. Mice in which GHRH neurons have been ablated using a novel viral ion channel transgene (GHRH-M2 mice) show marked anterior pituitary hypoplasia and GH deficiency although GH cells are present. GHRH-M2 mice are also deficient in prolactin which is surprising as GHRH has little or no direct effect on PRL synthesis or release (Le Tissier, Mol Endo 19). In mic...

Regulation of anterior pituitary Anx-1 during the estrus cycle of the rat

Davies E , Christian H , Buckingham J , Morris J

Annexin-1 (Anx-1), a 37 kDa protein, is a member of the super-family of Ca2+and phospholipid-binding annexin proteins and acts as a mediator of glucocorticoid (GC) action in the host defence and neuroendocrine systems. In the anterior pituitary Anx-1 is expressed mainly by folliculostellate (FS) cells and mediates the early-delayed feedback inhibition exerted by GCs on the release of ACTH and other pituitary hormones. The stress responsiveness ...

Does Annexin-1 have a biological function in the adrenal gland?

VEGF secretion from pituitary folliculo-stellate cells: a role for KATP channels?

Budzik JM , Christian HC , Omer S , Morris JF

VEGF (vascular endothelial growth factor) is an endothelial cell mitogen responsible for physiological and pathophysiological angiogenesis. Abnormal regulation of VEGF expression in anterior pituitary folliculostellate (FS) cells has been implicated in pituitary tumour progression. PACAP is a well-documented stimulant of VEGF secretion from the pituitary in which both FS and endocrine cells express VEGF. TtT/GF cells are a mouse pituitary cell line with many characteristics of...

Gonadotrophs in annexin 1 knockout mice demonstrate enhanced secretory activity by structural analysis

Wain S , Morris J , Buckingham J , Christian H

Annexin 1 (ANXA1) is a member of the annexin family of phospholipid- and calcium- binding proteins with a well-demonstrated role in the inhibitory actions of glucocorticoids on the release of ACTH, TSH, GH and PRL from the anterior pituitary. ANXA1 and ANXA1 truncated peptides have also recently been found to mimic glucocorticoid suppression of LH release (John et al, 2003 J Neuroendocrinol 15, 946-57). Here we test the hypothesis that because ANXA1 mediates some of the inhibi...

Comparison of anterior pituitary annexin 1 expression in male/female and old/young Long Evans and Brattleboro rats

Davies E , Christian H , Buckingham J , Morris J

It has been noted that there are differences in the hypothalamic-pituitary-adrenal (HPA) axis between the sexes, as well as changes in the axis with age. The differences in the HPA axis between the gender and age groups are thought to be due to differences in the efficiency of glucocorticoid (GC) negative feedback. The pituitary, a principle site of GC negative feedback, is the focus of this investigation. Annexin-1 (ANXA1) is a member of the annexin family of phospholipid- an...

Sexual dimorphism in anterior pituitary function in the annexin 1 knockout mouse

John C , Morris J , Christian H , Flower R , Buckingham J

Immunoneutralisation and antisense studies have shown that the acute regulatory actions of glucocorticoids (GCs) on the secretion of corticotrophin (ACTH) from the anterior pituitary gland are dependent upon the 37 kDa protein, annexin 1 (Anxa1 [1]), which is localised mainly to the S100-positive folliculo-stellate (FS) cells that lie in close apposition with the endocrine cells. The aim of this study was to examine pituitary function in male and female mice lacking the Anxa1 ...
